This first step can be performed in many ways, but the most common is performed using sodium dodecyl sulfate-polyacrylamide (SDS-PAGE) gel electrophoresis.

SDS is a detergent that denatures or degrades proteins by unfolding them and giving them an overall negative charge. After the proteins are denatured, they can be separated by gel electrophoresis.

Transfer proteins across the membrane

This can be achieved in many ways but has traditionally been achieved by wet transfer. The filter paper, gel, and nylon membrane are placed in a pile with each piece in contact with each other.

The stack order from bottom to top is as follows: gel, filter paper, nylon membrane, and another piece of filter paper. The bottom of the pile gets wetter and drier towards the top. For the transfer to take place, the nylon membrane must be placed directly over the gel.
